"The memorable art-deco image of Maria as a robot is based on a robot created in the laboratory of the city’s mad scientist, to resemble the dead wife of the city’s ruler. "

The scientist, "Rotwang", kidnaps Maria and decides to make a lifelike robot duplicate of her, a "gynoid", who will obey him. He's working for those who do not like a pro-worker, religious cult that the real Maria has started and wants to use the bizarre behavior of the robot Maria to destroy the real Maria's image of moral purity.

The real Maria is not killed by the replication process. She appears to pass out in the scene, but that might be because the device deep scanned her brain's memories and personality to make the robot of her.

However, the current versions of this film we have are actually made from bits and pieces salvaged over the years and portions of the original are still missing. In one of those missing portions it is mentioned that Rotwang actually secretly loved Maria, but she did not return his affection. It's possible he also wanted a robot Maria so he could order her love him.

Most people don't realize that about 80% of all of the audio and film material created before about 1930 has been permanently lost because of the fragile media it was recorded on that just deteriorated away unless carefully stored which was not done in most cases.

This film almost became part of that lost material which would have been a tragedy because it's such an important part of the history of sci-fi movies. For all we know there could be other similar films that were lost.
